One-Step Bacteria 16S  Library prep kit is a 16S sequencing library construction kit developed for Illumina sequencing platform. The kit adopts the optimized dual index amplification strategy to specifically amplify the v3-v4 region (319f-806r) of 16S rRNA gene of microbial sample. A complete sequencing library can be obtained by one-step PCR amplification, which can be directly used with Illumina & nbsp& nbsp; Pe300 sequencing was performed on miseq platform or PE250 sequencing was performed on hiseq2500 platform. The operation of the kit is simple, fast and flexible. The dual index strategy allows more samples to be sequenced.

Matters Needing Attention

1. Please read this manual carefully before the test and pay attention to the storage temperature of each component to ensure the successful completion of the test.

2. Please use high-quality DNA samples. Low quality samples may lead to the failure of database establishment.

3. Please pay attention to avoid cross contamination between nucleic acid samples and products during operation.

4. The magnetic beads used in the experiment should be taken out from the 4 ℃ environment 30 minutes in advance and balanced to room temperature. All magnetic bead operations shall be at room temperature.

5. 80% ethanol needs to be prepared and used now.

6. Materials to be prepared: absolute ethanol, RNase free water, 0.2ml PCR tube, 1.5ml centrifugal tube, magnetic frame, PCR instrument, etc;
